Influence of topiramate upon locomotor activity of phenobarbital in mice

Topiramate (TPM) is one of the newer antiepileptic drugs with fully unknown mechanism of action. We decided to evaluate the effects of TPM on locomotor activity of phenobarbital (PB) in mice. The effects of PB alone or in combination with TPM were studied on exploratory and spontaneous locomotor activity. In the present study, we showed that TPM (5 mg/kg) tested alone or co-administered with PB did not impair the performance of mice evaluated in three variables of exploratory and spontaneous locomotor activity (number of horizontal and vertical movements, average distance per movement). In contrast, PB (17.7 mg/kg) alone raised locomotor activity. The obtained results and previous findings, indicate that rational polytherapy with low doses of TPM did not induce behavioral side effects. It may conclude that the observed interaction may have a potential clinical significance.
