Investigation of triterpenic acids by TLC method in reversed-phase system

Abstract

Oleanolic, ursolic and betulinic acids belong to pentacyclic triterpenes  occurring widely in medicinal herbs and plants.  In the present work the applicability of TLC in reversed phase system for the separation of these triterpenes was studied. TLC was performed on HPTLC RP-18s and RP-18w plates. The mixtures of various solvenst eg.  water with: acetone, methanol, acetonitrile and tetrahydrofuran or mixtures of acetonitrile, methanol, isopropanol, diisopropyl ether, ethanol, ethyl acetate in various ratios, were tested. The best chromatographic system was used for densitometric  determination of betulinic acid in fresh birch bark. The densitograms were obtained using Desaga CD-60 densitometer (Heidelberg, Germany) controlled by a Pentium computer in absorbance/reflectance mode at λ = 530 nm. The obtained results was checked on HPLC.
